چکیده

THE main pathway of cholesterol catabolism involves the conversion of cholesterol to bile acids which are eliminated via the bile and feces. As liver cholesterol is considered to exist in equilibrium with serum cholesterol, nutritional factors accelerating cholesterol catabolism may lower the serum cholesterol level. Furthermore, it is well known that hypercholesterolemia and subsequent experimental atherosclerosis can be more easily produced in chickens than in rats; the latter are extremely resistant to experimental atherosclerosis. Only extensive treatment with dietary cholesterol, thiouracil, and cholic acid causes hyperlipemia in rats.'

In vivo metabolism-based discovery of a potent cholesterol absorption inhibitor, SCH58235, in the rat and rhesus monkey through the identification of the active metabolites of SCH48461.

SCH48461 is a selective and highly potent inhibitor of cholesterol absorption. In rats, SCH48461 is rapidly and completely metabolized in the first pass through the body. To compare the activity of the metabolites of SCH48461 with SCH48461 itself, an intestinally cannulated, bile duct-cannulated rat model for cholesterol absorption was developed. SCH48461 inhibited the absorption of cholesterol...

Metabolism in vivo of all-trans-retinoic acid. Biosynthesis of 13-cis-retinoic acid and all-trans- and 13-cis-retinoyl glucuronides in the intestinal mucosa of the rat.

The metabolites of all-trans-[3H]retinoic acid appearing in the intestines of bile duct-cannulated rats were compared to those of similarly treated intact rats. 2.4% of administered radioactivity was found in the small intestines of bile duct-cannulated rats 2 H after dose, while a much larger proportion of the dose (7.2%) was found in the intestines of the intact rats. All-trans- and 3-cis-ret...

Regulation of cholesterol and bile acid homoeostasis in bile-obstructed rats.

We examined how total blockage of biliary excretion, the major pathway through which cholesterol and bile acids are removed from the body, affects liver function, cholesterol and bile acid metabolism and homoeostasis. After 4 weeks of bile-duct ligation, rats showed impaired liver function, as documented by elevations in serum bilirubin and alkaline phosphatase activity. Moreover, bile-duct lig...

Effects of bile salts on glucose metabolism by slices of hamster small intestine.

Bile salts are known to be essential for the effective absorption of fat from the intestinal tract (1). They have been shown to stimulate the action of pancreatic lipase (2) and have an important emulsifying action on water-insoluble dietary triglycerides that is probably related to their ability to form micelles with fatty acids and monoglycerides (3). In addition to their role in triglyceride...
